Anti-Inflammatory and Anti-Arthritic Efficacies of an Indian Traditional Herbo-Mineral Medicine "Divya Amvatari Ras" in Collagen Antibody-Induced Arthritis (CAIA) Mouse Model Through Modulation of IL-6/IL-1β/TNF-α/NFκB Signaling.

Divya Amvatari Race (DAR), Herbo-mineral formulations Ayurvedic India, has been described in ancient texts to provide relief from inflammation of RA-related distress. Therefore, in this study, we explored the biocompatibility, anti-inflammatory and anti-arthritic efficacy of DAR using in vivo and in vitro models of disease. Using carrageenan (CA) -stimulated Wistar rat foot edema model, we showed a decrease in inflammation induced foot edema in a human equivalent dose of DAR.

CAIA onset of RA in mice was determined by using parameters such as improvement of arthritis score, and the induction of disease-related lesions in the leg and knee joints, and improved mechanical and thermal hyperalgesia. CAIA animal treatment with human equivalent dose significantly reversed DAR-related pathogenesis of RA. 

This effect is comparable to the standard drug treatment Caprine Clia Kits of RA, MTX. In lipopolysaccharide stimulated THP-1 cells, DAR was found to inhibit the release of IL-6, IL-1β, TNF-α and inflammatory gene upstream regulatory proteins, NF. This study supports the activities of anti-arthritic and anti-inflammatory medicine traditional Indian Herbo-mineral, DAR. These results also confirm that the DAR is very biocompatible and will show minimal health-related side effects than those associated with MTX standard of care. Taken together, we show that DAR can be used as an alternative or complementary therapy promises to treat rheumatoid arthritis
